Indications
- Treatment of premature ejaculation in men aged 18-64
- Persistent or recurrent ejaculation with minimal sexual stimulation
- Marked personal distress or interpersonal difficulty
- Poor control over ejaculation
Pharmacology
- Inhibition of neuronal reuptake of serotonin
- Sympathetic nervous system mediation of human ejaculation
- Rapid absorption with maximum plasma concentrations
- Metabolism to multiple metabolites
- Evidence of presystemic first-pass metabolism
- Rapid distribution with a mean steady state volume of distribution
- Terminal half life of approximately 19 hours
Dosage & Administration
- Recommended starting dose for adult men (18-64 years) is 30 mg
- Maximum recommended dosing frequency is once every 24 hours
- Recommended to be taken approximately 1 to 3 hours prior to sexual activity
- Not recommended for children, adolescents, or elderly (65 years and over)
- Caution advised in patients with mild or moderate renal impairment
- Contraindicated in patients with moderate and severe hepatic impairment
Interaction
- Caution advised for concomitant use with CNS active medicinal products
- Prescribed with caution in patients who use PDE5 inhibitors
- Recommended to be prescribed with caution in patients who use alpha adrenergic receptor antagonists
- Caution advised when used in patients taking Warfarin chronically
- Concomitant use of alcohol could increase the chance or severity of adverse reactions
Contraindications
- Known hypersensitivity to Dapoxetine Hydrochloride
- Significant pathological cardiac conditions
- Concomitant treatment with monoamine oxidase inhibitors (MAOIs) or thioridazine
- Concomitant treatment with serotonin reuptake inhibitors or medicinal/herbal products with serotonergic effects
