Pharmacology
- Binds to GABA receptor
- Potentiates effect of GABA
- Opens chloride ion channels into cells
- Rapid and complete absorption after oral administration
- Peak plasma concentration within 15-90 minutes
- Mean plasma half-life of 30 hours
- Metabolized in the liver
- Only traces of unchanged drug excreted in urine
- Eliminated with the feces
- Rectal administration is significantly absorbed
Dosage & Administration
- Oral dosage for anxiety and insomnia
- IM/slow IV injection for severe acute anxiety and febrile convulsion in children
- Rectal administration for children
- Dosage adjustment for elderly or debilitated patients
Interaction
- Contraindicated with alcohol
- Potential for increased sedation with neuroleptics, hypnotics, sedative antihistamines, and CNS depressants
- Drug clearance affected by concomitant administration of phenobarbitone and cimetidine
- Inhibition of metabolism by omeprazole and isoniazide
