Indications
- Treatment of endometriosis
- Reduces pelvic pain in women with endometriosis
- Reduces effects of estrogen on tissues
- Reduces growth effect of estrogen on endometrium and the breast
Pharmacology
- Acts as agonist at the progesterone receptor with weak affinity
- Causes endometrial atrophy after prolonged use
- Promotes antiproliferative, immunologic and antiangiogenic effects on endometrial tissue
- Reduces endogenous production of estradiol
- Antagonist at androgen receptors
- Improves androgenic symptoms such as acne and hirsutism
Dosage & Administration
- 2 mg tablet to be taken daily without any break
- Tablet-taking can be started on any day of the menstrual cycle
- Continuous administration without regard to vaginal bleeding
- In the event of missed tablet(s), take 2 mg as soon as remembered
Interaction
- Metabolized mainly by the cytochrome P450 3A4 system
- Known CYP3A4 inhibitors may increase plasma levels of progestogens
Contraindications
- Hypersensitivity to Dienogest or any excipients
- Active venous thromboembolic disorder
- Arterial and cardiovascular disease
- Diabetes mellitus with vascular involvement
- Severe hepatic disease
- Liver tumors (benign or malignant)
- Sex hormone-dependent malignancies
- Undiagnosed vaginal bleeding

- Weight increase
- Irritability
Pregnancy & Lactation
- Limited data from use in pregnant women
- No special risks on pregnancy, embryonic/fetal development, birth or development after birth for humans
- Not recommended during lactation
Precautions & Warnings
- Pregnancy must be excluded before starting treatment
- Patients advised to use nonhormonal methods of contraception
- Affects menstrual bleeding pattern
- Contraindicated in patients with severe hepatic disease
- Ovulation inhibited in the majority of patients
Use in Special Populations
- Not indicated in children prior to menarche
- Safety and efficacy in adolescents not established
- No relevant indication for use in geriatric population
Overdose Effects
- No risk of acute adverse effects in case of inadvertent intake of a multiple of the daily therapeutic dose
- No specific antidote
