Indications
- Relief from chronic heartburn symptoms
- Healing of erosive esophagitis
- Maintenance of healing of erosive esophagitis
- Eradication of Helicobacter pylori infection
- Zollinger-Ellison Syndrome
- Acid related Dyspepsia
- Duodenal & Gastric ulcer
Pharmacology
- Proton pump inhibitor
- Suppression of gastric acid secretion
- Absorption peak at approximately 1.5 hours
- Distribution: 97% bound to plasma proteins
- Metabolized in the liver by the cytochrome P450 (CYP) enzyme system
- Plasma elimination half-life of approximately 1–1.5 hours
Dosage
- 20 mg or 40 mg once daily for healing of erosive esophagitis for 4-8 weeks
- 20 mg once daily for maintenance of healing of erosive esophagitis
- 20 mg once daily for symptomatic GERD for 4 weeks
- Triple therapy for Helicobacter pylori eradication
- 20-80 mg once daily for Zollinger-Ellison Syndrome
- 20-40 mg once daily for Acid-related Dyspepsia
- 20 mg once daily for Duodenal ulcer for 2-4 weeks
- 20-40 mg once daily for Gastric ulcer for 4-8 weeks
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Oral Suspension: Should be stirred well and consumed within 30 minutes. If administered through a tube, use an appropriately sized syringe
- IV Injection: Administered intravenously over a period of at least 3 minutes
Interaction
- Extensively metabolized in the liver by CYP2C19 and CYP3A4
- Potentially interferes with the absorption of drugs where gastric pH is an important determinant of bioavailability
- No clinically significant interactions with certain drugs
Contraindications
- Contraindicated in patients with known hypersensitivity to any of the formulation

Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Excretion in milk not studied
- Breast-feeding should be discontinued if essential
Precautions & Warnings
- Symptomatic response does not preclude the presence of gastric malignancy
- Tablet should be taken at least one hour before meals
- Antacids may be used while taking the medicine
Use in Special Populations
- Safety and effectiveness in paediatric patients not established
- No overall differences in safety and efficacy observed between the elderly and younger individuals
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
- Pharmacokinetics not expected to be altered in renal impairment
Overdose Effects
- A single oral dose at 510 mg/kg has been lethal to rats
- No specific antidote known
- Treatment should be symptomatic and supportive

- Store at a temperature not exceeding 30°C in a dry place
