Indications
- Chronic management of hyperuricemia
- Indicated for gout patients
Pharmacology
- Non-purine, selective xanthine oxidase (XO) inhibitor
- Decreases serum uric acid level
- Inhibits xanthine oxidase
- Not expected to inhibit other enzymes involved in purine and pyrimidine synthesis
Dosage & Administration
- Recommended starting dose of 40 mg tablet once daily
- Dose adjustment based on serum uric acid level
- Dosage for tumor lysis syndrome
- Gout flare management instructions
- No dose adjustment needed for renal impairment
- No dose adjustment needed for mild to moderate hepatic impairment
- Not established for pediatric use
Interaction
- Contraindicated with mercaptopurine/azathioprine
- No dose adjustment required with rosiglitazone/CYP2C8 substrates
- Metabolism depends on Uridine Glucuronosyl Transferase (UGT) enzymes
- Monitoring recommended for potent inducers of UGT enzymes
- Can be co-administered with colchicine/indometacin/hydrochlorothiazide/warfarin
Contraindications
- Contraindicated in patients being treated with azathioprine, mercaptopurine, or theophylline
