Indications
- Irritable Bowel Syndrome with Constipation (IBS-C)
- Chronic Idiopathic Constipation (CIC) in adults
- Opioid-Induced Constipation (OIC) in adult patients with chronic, non-cancer pain
Pharmacology
- Locally-acting chloride-channel activator
- Increases intestinal secretion
- Increases intestinal motility
- Activates ClC-2 chloride channels
- Causes efflux of chloride ions
- Leads to efflux of sodium ions
- Stimulates recovery of muscosal barrier function
Dosage & Administration
- Recommended dose for IBS-C is 8 mcg twice daily with food and water
- Recommended dose for Chronic Idiopathic Constipation and OIC is 24 mcg twice daily with food and water
- Dose adjustment for hepatic impairment
Interaction
Possibility of a dose-dependent decrease in efficacy with diphenylheptane opioids
Contraindications
Contraindicated in patients with known or suspected mechanical gastrointestinal obstruction
