Indications

  • Relief from chronic heartburn symptoms
  • Healing of erosive esophagitis
  • Maintenance of healing of erosive esophagitis
  • Eradication of Helicobacter pylori infection
  • Treatment of Zollinger-Ellison Syndrome
  • Management of Acid-related Dyspepsia
  • Treatment of Duodenal & Gastric ulcer

Pharmacology

  • Proton pump inhibitor
  • Suppression of gastric acid secretion
  • H+/K+-ATPase inhibition
  • Enteric-coated pellet formulation
  • Peak plasma levels at approximately 1.5 hours
  • Metabolized in the liver by CYP enzyme system

Dosage

  • Healing of Erosive Esophagitis: 20mg or 40mg Once Daily for 4-8 Weeks
  • Symptomatic GERD: 20mg Once Daily for 4 Weeks
  • Helicobacter Pylori eradication: Triple Therapy for 10 days
  • Zollinger-Ellison Syndrome: 20-80mg Once Daily
  • Acid-related Dyspepsia: 20-40mg Once Daily for 2-4 weeks

Administration

  • Swallowed whole and taken one hour before a meal
  • Oral suspension instructions
  • IV Injection/Infusion guidelines

Interaction

  • Metabolized by CYP2C19 and CYP3A4
  • Potential interference with CYP2C19
  • No clinically significant interactions with specific drugs

Contraindications

  • Contradicted in patients with known hypersensitivity to any of the formulation

Pregnancy lactation

  • Symptomatic response does not rule out gastric malignancy
  • Instructions for difficult swallowing patients
  • Use of antacids while taking Esoral

Special populations

  • No established safety and effectiveness in pediatric patients
  • No overall differences in safety and efficacy in elderly individuals
  • No recommended dosage adjustment for mild to moderate hepatic insufficiency
  • No expected alteration in pharmacokinetics with renal impairment

Overdose effects

  • Lethal at high oral doses in rats
  • Signs of acute toxicity include reduced motor activity and convulsions
  • No specific antidote known
