Indications
- Used in adult type 2 diabetes Mellitus
- When diet, exercise, and weight loss alone are not effective in controlling blood sugar
Pharmacology
- Second generation sulfonylurea drug
- Stimulates insulin release from pancreatic cells
- Facilitates Ca+2 transport across beta-cell membranes
- Decreases hepatic glucose output
Dosage & Administration
- Usual initial dose: 40-80mg daily
- Dose can be increased up to 320mg daily in divided doses
- Modified release preparation available
- Recommended daily dose: 1 to 4 tablets (maximum 120 mg)
- To be taken with a glass of water at breakfast time
- Swallow whole, do not chew or crush
- Always eat a meal after taking the tablet
Interaction
- May interact with other oral antidiabetics, GLP-1 receptor agonists, insulin, antibiotics, medicines for high blood pressure, heart failure, fungal infections, ulcers, depression, painkillers, and more
- May increase effects of medicine that reduce blood clotting
- May be affected by alcohol consumption
- May impair ability to concentrate or react, especially under low or high blood sugar conditions
Contraindications
- Allergic to gliclazide or related medicines
- Insulin-dependent diabetes (type 1)
- Ketone bodies and sugar in urine
- Severe kidney or liver disease
- Taking medicines for fungal infections
- Breastfeeding

- Most common: low blood sugar (hypoglycemia)
- May cause liver disorders, skin disorders, blood disorders, digestive disorders, and eye disorders
- Rare: severe changes in blood cells, allergic inflammation of blood vessel wall, reduction in blood sodium, symptoms of liver impairment
