Description
- Indicated as an adjunct to diet and exercise for lowering blood glucose in noninsulin dependent (Type II) diabetes mellitus patients
- Stimulates insulin release from pancreatic beta cells
- Reduces basal hepatic glucose production
- Increases peripheral tissue sensitivity to insulin
- Decreases blood glucose concentration
- Prescribed based on regular blood sugar monitoring
- Tablet to be swallowed with liquid
- Not suitable for insulin dependent (type I) diabetes mellitus
- Must not be taken during pregnancy or while breastfeeding
- Hypoglycaemia may be increased in initial weeks of treatment
Dosage
- Initial dose: 1 mg once daily
- Usual dose range: 1 to 4 mg daily
- Timing and distribution decided by physician
- Dose adjustment based on diabetes control and individual factors
- Changeover guidance from other oral antidiabetics
Storage Conditions
- Do not store above 30°C
- Keep away from light and out of the reach of children
Pharmacology
- Stimulates insulin release from pancreatic beta cells
- Improves sensitivity of beta cells to glucose stimulus
- Reduces basal hepatic glucose production
- Increases peripheral tissue sensitivity to insulin
- Hypoglycaemic action persists for 24 hours in non-fasting diabetic patients
Interaction
- Potentiating drugs include ACE inhibitors, male sex hormones, chloramphenicol, fluconazole, sulphonamides
- Attenuating drugs include barbiturates, corticosteroids, diazoxide, diuretics
Contraindications
- Not suitable for insulin dependent (type I) diabetes mellitus
- Not for diabetic ketoacidosis or diabetic coma
- Avoid in patients hypersensitive to Glimepiride or sulfonylureas
- Avoid in severe hepatic dysfunction and severe impairment of renal function

- Abdominal pain
- Urticaria
- Fall in blood pressure
Pregnancy & Lactation
- Not to be taken during pregnancy
- Patients planning a pregnancy should change over to insulin
- Not to be taken by breastfeeding women
Precautions & Warnings
- Initial weeks of treatment may increase risk of hypoglycaemia
- Hypoglycaemia can be promptly controlled by immediate intake of carbohydrates
Overdose Effects
- Overdosage can produce hypoglycaemia
- Mild hypoglycaemic symptoms should be treated with oral glucose
- Severe hypoglycaemic reactions constitute medical emergencies
