Indications
- Adjunct to diet and exercise for noninsulin dependent diabetes mellitus (NIDDM)
- Combination use with metformin
- Combination use with insulin
Pharmacology
- Stimulates insulin release from pancreatic beta cells
- Reduces basal hepatic glucose production
- Increases peripheral tissue sensitivity to insulin
Dosage
- Initial dose is 1 mg once daily
- Dose titration based on regular blood sugar monitoring
- Usual dose range for well-controlled diabetes is 1 to 4 mg daily
Administration
- Must be swallowed with sufficient liquid
Interaction
- Potentiates hypoglycaemic action with ACE inhibitors
- Attenuates hypoglycaemic action with barbiturates and corticosteroids
- May mask hypoglycaemic symptoms when used with beta-receptor blockers
Contraindications
- Not suitable for insulin-dependent diabetes mellitus
- Not for use in diabetic ketoacidosis or diabetic coma
- Contraindicated in patients hypersensitive to sulfonylureas

Pregnancy & Lactation
- Not to be taken during pregnancy
- Changeover to insulin required
- Not to be taken while breastfeeding
Precautions & Warnings
- Initial weeks of treatment may increase risk of hypoglycaemia
- Immediate intake of carbohydrates for controlling hypoglycaemia
Overdose Effects
- Overdosage can produce hypoglycaemia
- Mild symptoms should be treated with oral glucose and dosage adjustments
- Severe hypoglycaemic reactions require immediate hospitalization
