Indications
- Adjunct to diet and exercise to lower blood glucose in noninsulin-dependent diabetes mellitus (NIDDM)
- Use in combination with metformin or insulin for adequate glycaemic control
Pharmacology
Stimulates insulin release from pancreatic beta cells, reduces hepatic glucose production, improves peripheral tissue sensitivity to insulin
Dosage
- Initial Dose: 1 mg once daily
- Usual Dose Range: 1 to 4 mg daily
- Timing and Distribution: Decided by physician based on patient's lifestyle
- Secondary Dosage Adjustment: Considered as control of diabetes improves
- Changeover from Other Oral Antidiabetics: Initial dose of 1 mg
Administration
Tablets must be swallowed with sufficient liquid
Interaction
- Potentiate the Hypoglycaemic Action:
- ACE inhibitors
- Cyclophosphamide
- Fluconazole
- Attenuate the Hypoglycaemic Action:
- Acetazolamide
- Barbiturates
- Calcium channel blockers
Contraindications
- Insulin dependent diabetes mellitus
- Diabetic ketoacidosis
- Hypersensitivity to Glimepiride or sulfonylureas
- Severe hepatic dysfunction
- Severe impairment of renal function
Side Effects
- Hypoglycaemia
- Nausea
- Temporary visual impairment
- Abdominal pain
Pregnancy & Lactation
Not suitable for use during pregnancy or breastfeeding
Precautions & Warnings
Increased risk of hypoglycaemia in initial weeks of treatment, necessitates careful monitoring
Overdose Effects
Can produce hypoglycaemia, requiring aggressive treatment with glucose and close monitoring
