Pharmacology
- Proton pump inhibitor
- Suppression of gastric acid secretion

- Peak plasma levels occur at approximately 1.5 hours
- Systemic bioavailability is approximately 90%
Dosage
- Oral dosage form
- Healing of erosive esophagitis: 20 mg or 40 mg Once Daily for 4-8 Weeks
- Maintenance of healing of erosive esophagitis: 20 mg Once Daily
- Symptomatic GERD: 20 mg Once Daily for 4 Weeks
- Helicobacter Pylori eradication: Triple Therapy
- Zollinger-Ellison Syndrome: 20-80 mg once daily
- Acid-related Dyspepsia: 20-40 mg once daily
- Duodenal ulcer: 20 mg once daily for 2-4 weeks
- Gastric ulcer: 20-40 mg once daily for 4-8 weeks
- IV Injection or IV infusion for various conditions and patient age groups
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal

Interaction
- Extensive metabolism in the liver by CYP2C19 and CYP3A4
- Inhibition of gastric acid secretion
- Potential interference with CYP2C19
- No clinically significant interactions with various drugs

- Known hypersensitivity to any of the formulation

- Specific instructions for patients with difficulty swallowing capsules
- Use of antacids while taking Nexe
Use in Special Populations
- Safety and effectiveness in paediatric patients not established
- No overall differences in safety and efficacy in the elderly
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
- No alteration of pharmacokinetics in patients with renal impairment
Overdose Effects
- Lethal to rats at a high dose
- Signs of acute toxicity
- No specific antidote known
- Symptomatic and supportive treatment recommended
