Mode of Action
- Proton pump inhibitor
- Suppresses gastric acid secretion
- Specific inhibition of the H+/K+-ATPase in the gastric parietal cell
Absorption
- Peak plasma levels occur at approximately 1.5 hours after oral administration
Distribution
- 97% bound to plasma proteins
- Constant plasma protein binding over the concentration range
Metabolism
- Extensively metabolized in the liver by the cytochrome P450 (CYP) enzyme system
Excretion
- Plasma elimination half-life of approximately 1–1.5 hours
- Less than 1% excreted unchanged in the urine

Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Suspension should be consumed within 30 minutes
- IV Injection must be administrated intravenously over a period of at least 3 minutes
Interaction
- Extensively metabolized in the liver by CYP2C19 and CYP3A4
- Not likely to inhibit CYPs 1A2, 2A6, 2C9, 2D6, 2E1, and 3A4
- No clinically significant interactions with certain drugs
Contraindications
- Known hypersensitivity to any of the formulation

- Patients should take capsules at least one hour before meals
- Antacids may be used while taking Progut

- Geriatric Use: No overall differences in safety and efficacy observed between elderly and younger individuals
Overdose Effects
- Reduced motor activity
- Changes in respiratory frequency
- Tremor
- Ataxia
- Intermittent clonic convulsions
- No specific antidote known
- Treatment should be symptomatic and supportive
