Description

  • A proton pump inhibitor used to relieve chronic heartburn symptoms and GERD
  • Indicated for the healing of erosive esophagitis and maintenance of healing
  • Used in combination with amoxicillin and clarithromycin for eradication of Helicobacter pylori infection
  • Treats Zollinger-Ellison Syndrome, acid-related Dyspepsia, duodenal & gastric ulcers
  • Suppresses gastric acid secretion by inhibiting H+/K+-ATPase in the gastric parietal cell
  • Enteric-coated pellet formulation of esomeprazole magnesium for better acid control
  • Absorption peak plasma levels occur at approximately 1.5 hours after oral administration
  • Distribution: 97% bound to plasma proteins, apparent volume of distribution at steady state is approximately 16 L
  • Extensively metabolized in the liver by the cytochrome P450 (CYP) enzyme system
  • Plasma elimination half-life is approximately 1–1.5 hours, less than 1% of parent drug is excreted in the urine
  • Adult dosage: 20 mg or 40 mg once daily for healing of erosive esophagitis and symptomatic GERD
  • Triple therapy for Helicobacter pylori eradication: 40 mg Esomeprazole once daily for 10 days
  • Dosages for Zollinger-Ellison Syndrome, acid-related Dyspepsia, duodenal and gastric ulcers
  • Oral administration at least one hour before meals, IV injection duration, and infusion time
  • No clinically relevant interactions with drugs metabolized by CYPs 1A2, 2A6, 2C9, 2D6, 2E1 and 3A4
  • Potential interference with CYP2C19, absorption of drugs affected by gastric pH
  • Storage at a temperature not exceeding 30°C, protected from light and moisture
