Pharmacology

  • Proton pump inhibitor that suppresses gastric acid secretion
  • Single optical isomer providing better acid control
  • Absorbed after oral administration
  • Extensively metabolized in the liver
  • Plasma elimination half-life of approximately 1–1.5 hours

Dosage

  • 20 mg or 40 mg Once Daily for 4-8 Weeks for healing of erosive esophagitis
  • 20 mg Once Daily for maintenance of healing of erosive esophagitis
  • 20 mg Once Daily for symptomatic GERD
  • 40 mg Once Daily for Helicobacter Pylori eradication
  • 20-80 mg once daily for Zollinger-Ellison Syndrome

Administration

  • Tablet or capsule should be swallowed whole and taken one hour before a meal
  • Oral Suspension should be mixed with water and consumed within 30 minutes
  • IV Injection or IV infusion to be administered as per specific protocols

Interaction

  • Extensively metabolized in the liver by CYP2C19 and CYP3A4
  • No clinically relevant interactions with specific drugs metabolized by certain CYP enzymes expected
  • Potential interference with CYP2C19
  • Combination Therapy with Clarithromycin can influence plasma levels

Contraindications

  • Contraindicated in patients with known hypersensitivity to any of the formulation

  • Specific information for patients on capsule consumption
  • Use of antacids

  • No overall differences in safety and efficacy observed between the elderly and younger individuals
  • No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
