Indications

  • Relief from chronic heartburn symptoms
  • Healing of erosive esophagitis
  • Maintenance of healing of erosive esophagitis
  • Eradication of Helicobacter pylori infection
  • Treatment of Zollinger-Ellison Syndrome
  • Management of acid-related Dyspepsia
  • Treatment of Duodenal & Gastric ulcers

Pharmacology

  • Mode of Action: Proton pump inhibitor
  • Absorption: Enteric-coated pellet formulation
  • Distribution: 97% bound to plasma proteins
  • Metabolism: Extensively metabolized in the liver
  • Excretion: Less than 1% excreted in the urine

Dosage

  • Oral Dosage Form:
    • Healing of Erosive Esophagitis: 20 mg or 40 mg Once Daily for 4-8 Weeks
    • Symptomatic GERD: 20 mg Once Daily for 4 Weeks
    • Zollinger-Ellison Syndrome: 20-80 mg once daily
    • Acid-related Dyspepsia: 20-40 mg once daily for 2-4 weeks
  • IV Injection or IV Infusion: Dosing varies based on patient age, weight, and specific condition

Administration

  • Tablet or capsule: Swallowed whole one hour before a meal
  • Oral Suspension:
    • Stirred with water and consumed within 30 minutes
    • Administered through a nasogastric or gastric tube
  • IV Injection: Intravenously over a period of at least 3 minutes

Interaction

  • Metabolism: Primarily by CYP2C19 and CYP3A4
  • Drug Interactions:
    • Not likely to inhibit CYPs 1A2, 2A6, 2C9, 2D6, 2E1, and 3A4
    • May interfere with the absorption of certain drugs
