Indications
- Short-term management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Decrease of eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Metabolized in the liver
- Products of metabolism and some unchanged drugs are excreted in the urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system
- Demonstrates minimal anti-inflammatory effect at its analgesic dose
Dosage & Administration
- Recommended tablet dose: 10 mg every 4-6 hours for short-term use (up to 7 days)
- Recommended injection dose for adults: Single or multiple doses of 30-60 mg for moderate to severe acute pain
- Onset of analgesic effect within 30 minutes for both IV and IM administration
- Duration of analgesic effect: 4 to 6 hours
- Recommended eye drops dose for adults: 1 drop in each eye 4 times daily
Interactions
- Increased side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduced anti-hypertensive effect with beta blockers
- Increased risk of renal impairment with ACE inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to the drug or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic use before surgery

Precautions & Warnings
- Exercise caution in patients over the age of 65
- Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, or liver dysfunction
- Discontinue use of topical NSAIDs in patients with evidence of corneal epithelial breakdown
- Close monitoring for corneal health in such patients
- Avoid administration while wearing contact lenses for ophthalmic solution
