Indications

Description

  • Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Considered as a peripherally acting analgesic
  • Biological activity associated with the S-form
  • Linear pharmacokinetic property
  • Highly protein bound
  • Metabolized in the liver
  • Excretion in the urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibits cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet recommended dose of 10 mg every 4-6 hours for up to 7 days
  • Injection for moderately severe, acute pain management
  • IV bolus administration over no less than 15 seconds
  • IM administration to be given slowly and deeply into the muscle
  • Onset of analgesic effect within 30 minutes with maximum effect in 1 to 2 hours
  • Duration of analgesic effect is usually 4 to 6 hours
  • Different dosing regimens based on age, renal function, and body weight

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with beta blockers
  • Increased risk of renal impairment with ACE inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Children under 16 years of age
  • Prophylactic analgesic before surgery
