Indications
- Short-term management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Reduction of eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
- Active ingredient: Ketorolac Tromethamine
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Peripherally acting analgesic
- Pharmacokinetic property: linear
- Highly protein bound
- Metabolized in the liver
- Excreted in the urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits cyclooxygenase enzyme system
- Demonstrates minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours for short-term use (up to 7 days)
- Injection: Single or multiple doses for moderately severe, acute pain
- IV bolus must be given over no less than 15 seconds
- Analgesic effect begins within 30 minutes with maximum effect in 1 to 2 hours after dosing IV or IM
- Duration of analgesic effect is usually 4 to 6 hours
- Eye Drops: Adults: 1 drop in each eye 4 times daily
Interaction
- Other NSAIDs or aspirin: Increase the side effects of K-Lac
- Anti-coagulants: Enhance anti-coagulant effect
- Beta Blocker: Reduce the anti-hypertensive effect
- ACE Inhibitors: Increase the risk of renal impairment
- Methotrexate: Enhance the toxicity of methotrexate
Contraindications
- Hypersensitivity to this drug or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic before surgery

Precautions & Warnings

- Caution in patients with active or suspected peptic ulcer or gastrointestinal bleeding; asthma; liver dysfunction
- Discontinue use and monitor closely for corneal health in patients with evidence of corneal epithelial breakdown
