Indications
- Management of moderate to severe post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Decrease eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5-benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Peripherally acting analgesic
- Highly protein bound
- Metabolized in the liver
- Excretion in the urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours for short-term use (up to 7 days)
- Injection: Used for management of moderately severe, acute pain in postoperative setting
- Analgesic effect begins within 30 minutes for IV or IM administration
- Eye Drops: Adult dosage is 1 drop in each eye 4 times daily
Interaction
- Other NSAIDs or aspirin: Increases side effects of K-Lac
- Anti-coagulants: Enhances anti-coagulant effect
- Beta Blocker: Reduces anti-hypertensive effect
- ACE Inhibitors: Increases risk of renal impairment
- Methotrexate: Enhances toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Contraindicated in children under 16 years of age
- Contraindicated as prophylactic analgesic before surgery

Precautions & Warnings
- Caution in patients over 65 years
- Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, and liver dysfunction
- Discontinue use of topical NSAIDs with evidence of corneal epithelial breakdown
- Monitor corneal health in case of corneal epithelial breakdown
- Avoid administration while wearing contact lenses for ophthalmic solution
