Indications

Description

  • Pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Associated with the S-form
  • Linear pharmacokinetic property
  • Highly protein bound and largely metabolized in liver
  • Metabolites and unchanged drugs excreted in urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibits cyclooxygenase enzyme system and prostaglandin synthesis
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet dose: 10 mg every 4-6 hours for up to 7 days
  • Injection dose: varies based on age, weight, and condition
  • Duration of analgesic effect: 4 to 6 hours
  • Eye Drops dose: 1 drop in each eye 4 times daily

Interaction

  • Increase the side effects of Doloket with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with beta blockers
  • Increased risk of renal impairment with ACE inhibitors
  • Enhanced toxicity of methotrexate with Doloket

Contraindications

  • Hypersensitivity to the drug or other NSAIDs
  • Children under 16 years of age
  • Prophylactic analgesic before surgery
