Description
- Belongs to pyrrolo-pyrrole group of NSAIDs
- Chemically known as 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid
- Inhibits synthesis of prostaglandins
- Considered as a peripherally acting analgesic
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Metabolized in the liver
- Metabolites excreted in the urine
Pharmacology
- Potent analgesic NSAID
- Inhibits cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet recommended dose of 10 mg every 4-6 hours
- Injection recommended for moderate to severe acute pain management
- IV bolus administration over 15 seconds
- IM administration given slowly and deeply into the muscle
- Analgesic effect begins within 30 minutes
- Maximum effect in 1 to 2 hours after dosing IV or IM
- Duration of analgesic effect is 4 to 6 hours

Interaction
- Increased side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduced anti-hypertensive effect with beta blockers

- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac and other NSAIDs
- Not recommended for children under 16 years of age
- Contraindicated as prophylactic analgesic before surgery
