Description

  • Belongs to pyrrolo-pyrrole group of NSAIDs
  • Chemically known as 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid
  • Inhibits synthesis of prostaglandins
  • Considered as a peripherally acting analgesic
  • Biological activity associated with the S-form
  • Linear pharmacokinetic property
  • Highly protein bound
  • Metabolized in the liver
  • Metabolites excreted in the urine

Pharmacology

  • Potent analgesic NSAID
  • Inhibits cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet recommended dose of 10 mg every 4-6 hours
  • Injection recommended for moderate to severe acute pain management
  • IV bolus administration over 15 seconds
  • IM administration given slowly and deeply into the muscle
  • Analgesic effect begins within 30 minutes
  • Maximum effect in 1 to 2 hours after dosing IV or IM
  • Duration of analgesic effect is 4 to 6 hours

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants

Contraindications

  • Hypersensitivity to Ketorolac and other NSAIDs
  • Not recommended for children under 16 years of age
  • Contraindicated as prophylactic analgesic before surgery
