Indications
- Short-term management of moderate to severe acute post-operative pain

Description
- Pyrrolo-pyrrole group NSAID
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Metabolized in liver
- Metabolites and some unchanged drugs excreted in urine
Pharmacology
- Potent analgesic NSAID
- Inhibits cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose of 10 mg every 4-6 hours for short-term use (up to 7 days)
- Injection: Single or multiple doses for management of moderately severe acute pain, duration of analgesic effect usually 4 to 6 hours
- Eye Drops: Recommended dosage is 1 drop in each eye 4 times daily
Interaction
- Increased side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduction of anti-hypertensive effect with beta blockers
- Increased risk of renal impairment with ACE Inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to ketorolac or other NSAIDs
- Not for use in children under 16 years of age
- Contraindicated as prophylactic analgesic before surgery

- Discontinue use of topical NSAIDs in patients with corneal epithelial breakdown
