Indications
- Short-term management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Decrease eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Inhibits synthesis of prostaglandins
- Biological activity associated with the S-form
- Pharmacokinetic property is linear
- Highly protein bound
- Metabolized in the liver
- Products of metabolism and some unchanged drugs are excreted in the urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet recommended dose: 10 mg every 4-6 hours for up to 7 days
- Tablet not recommended for chronic use or doses exceeding 40 mg/day
- Injection may be used for moderately severe acute pain
- Analgesic effect begins within 30 minutes
- Maximum effect in 1 to 2 hours after dosing IV or IM
- Duration of analgesic effect is usually 4 to 6 hours
- Specific dosage recommendations for different age groups and conditions
Interaction
- Petorolac interaction with other NSAIDs or aspirin
- Enhanced effect with anti-coagulants
- Reduced anti-hypertensive effect with Beta Blocker
- Increased risk of renal impairment with ACE Inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to drug or other NSAIDs
- Not to be used in children under 16 years of age
- Contraindicated as prophylactic analgesic before surgery

Precautions & Warnings
- Caution in patients over the age of 65 years
- Caution in patients with active or suspected peptic ulcer
- Caution in patients with gastrointestinal bleeding or asthma
- Caution in patients with liver dysfunction
- Discontinue use of topical NSAIDs in presence of corneal epithelial breakdown
- Monitoring of corneal health for patients with corneal epithelial breakdown
- Avoid administering while wearing contact lenses for ophthalmic solution
