Indications
- Adjuvant treatment of postmenopausal women with hormone receptor positive early breast cancer
- Treatment of advanced breast cancer in women with postmenopausal status
- First-line treatment in postmenopausal women with hormone-dependent advanced breast cancer
- Pre-operative therapy in postmenopausal women with localized hormone receptor positive breast cancer
Pharmacology
- Potent and highly specific nonsteroidal aromatase inhibitor
- Inhibition of the aromatase enzyme
- Reduction of estrogen biosynthesis in all tissues
- Effects on estrogen biosynthesis in postmenopausal women
- Rapid and complete absorption from the gastrointestinal tract
- Plasma protein binding of approximately 60%
Dosage & Administration
- Recommended dose of 2.5 mg once daily
- Treatment duration of 5 years in certain cases
- No dose adjustment required for elderly patients or those with hepatic and/or renal impairment
Interaction
- No clinically significant interactions with cimetidine and warfarin
Contraindications
- Known or suspected hypersensitivity to letrozole or other aromatase inhibitors
- Contraindicated during pregnancy, lactation, and in pre-menopausal women
- Contraindicated in severe hepatic dysfunction
Side Effects
- Common side effects such as hot flushes, increased cholesterol levels, fatigue, pain in bones and joints, skin rash, headache, dizziness, gastrointestinal disorders
- Rare side effects including nervous disorders, memory problems, eye irritation, weight decrease, urinary tract infection
Pregnancy & Lactation
- Teratogenicity and maternal toxicity observed in pregnant rats
- Not recommended for use in pregnant women or nursing mothers
Precautions & Warnings
- No dosage adjustment necessary in breast cancer patients with moderate hepatic dysfunction
- Renal impairment did not affect plasma concentration at certain doses
- Potential risks of fatigue and dizziness should be considered
- Consideration for patients operating machinery or driving a car
