Indications
- Treatment and prophylaxis of mania
- Bipolar disorder
- Recurrent depression
Pharmacology
- Mechanism of action still unknown
- Inhibition of enzymes
- Modulation of glutamate receptors
- Alteration of intraneuronal metabolism
Dosage & Administration
- Initial adult dosage of 1 gm to 1.5 gm daily
- Prophylaxis initial dosage of 300-400 mg daily
- Should be taken with food
Interaction
- Reduced serum levels with certain inhibitors and preparations
- Enhanced hypothyroid effects with iodine salts
- Reduced pressor response to sympathomimetics
Contraindications
- Renal insufficiency
- Cardiovascular insufficiency
- Addison's disease
- Untreated hypothyroidism

- Tiredness
- Loss of appetite
- Nausea
- Vomiting
- Diarrhoea
- Hands shaking
- Memory problems
- Increased thirst
- Frequent urination
Pregnancy & Lactation
- Pregnancy Category D
- Increased incidence of cardiovascular abnormality in infants
- Breastfeeding not advised unless benefits outweigh the advantages
Precautions & Warnings
- Decreased tolerance from sweating or diarrhoea
- Supplemental fluid and salt administration under medical supervision
Use in Special Populations
- Renal Impairment: CrCl 10-50: 50-75% of normal dose
Overdose Effects
- Serum estimations every six hours
- No use of diuretics
- Osmotic diuresis or alkalinisation of the urine
- Peritoneal or haemodialysis if necessary
- Serum lithium levels monitoring for at least a week
