Indications

Pharmacology

  • Proton pump inhibitor
  • Suppression of acid secretion
  • Final step in gastric acid production
  • H+/K+ ATPase enzyme system
  • Inhibition of both basal and stimulated gastric acid secretion

Dosage & Administration

  • Benign gastric ulcer: 40 mg daily in the morning
  • Gastro-esophageal reflux disease: 20-40 mg daily
  • Duodenal ulcer: 40 mg daily in the morning
  • Duodenal ulcer associated with Helicobacter pylori: 40 mg twice daily
  • Prophylaxis of NSAID-associated gastric or duodenal ulcer: 20 mg daily
  • Zollinger-Ellison Syndrome: Initially 80 mg

Interaction

  • No significant drug interactions observed in clinical studies

Contraindications

  • Known hypersensitivity to any components of the formulation

Side Effects

  • Well-tolerated in both short-term and long-term treatment
  • Common side effects: headache, diarrhea
  • Rare side effects: abdominal pain, flatulence, rash, insomnia, hyperglycemia

Pregnancy & Lactation

  • US FDA Pregnancy Category B
  • No adequate and well-controlled studies in pregnant women
  • Excreted in human milk

Precautions & Warnings

  • Tablets should not be split, chewed, or crushed
  • Long-term therapy may lead to malabsorption of Vitamin B12
  • Increased risk of osteoporosis related disorders
