Indications
- Treatment of hypertension
- Reducing risk of stroke in patients with hypertension and left ventricular hypertrophy
Pharmacology
- Potent vasoconstrictor
- Affects renin-angiotensin system
- Stimulates aldosterone secretion
- Blocker of angiotensin II binding
Dosage
- Usual starting dose of 50/12.5 is one tablet once daily
- Maximum dose is 100/25 once daily
- Antihypertensive effect attained within three weeks after initiation
Administration
- May be administered with other antihypertensive agents
- Can be taken with or without food
Interaction
- No significant drug-drug interactions with Losartan Potassium
- Potential interactions with thiazide diuretics and antidiabetic drugs
- Additive effect with other antihypertensive drugs
Contraindications
- Hypersensitivity to any component of the product
- Contraindicated in patients with anuria or hypersensitivity to sulfonamide-derived drugs
Side Effects
- Symptomatic hypotension including dizziness
- Occasional hyperkalaemia
- Vertigo
- Gastro-intestinal disturbances
- Angioedema
- Headache
- Urticaria
- Pruritus
Pregnancy & Lactation
- Avoided in pregnancy unless essential
- Not recommended in breastfeeding
Precautions & Warnings
- Periodic determination of serum electrolytes
- Hypokalemia may rarely develop
- Impaired renal function should be monitored
- Symptomatic hypotension should be considered
Use in Special Populations
- Usual regimens may be followed in patients with creatinine clearance >30 ml/min
- Loop diuretics preferred in severe renal impairment
- Not recommended in patients with hepatic impairment
- Safety and effectiveness not established in pediatric patients
Overdose Effects
- Likely manifestations include hypotension, tachycardia, and bradycardia
- Supportive treatment should be instituted if symptomatic hypotension occurs
- Most common signs and symptoms involve electrolyte depletion
