Indications

Pharmacology

  • Broad-spectrum anthelmintic
  • Active against most nematodes
  • Inhibits formation of worms' microtubules
  • Causes worms' glucose depletion
  • 2-10% oral absorption
  • Peak plasma concentration within 30 minutes to 7 hours
  • Highly bound to plasma protein
  • Elimination half-life of 2.8 to 9 hours

Dosage & Administration

  • Adult dosage: 100 mg single dose for threadworms
  • Adult dosage: 100 mg twice daily for 3 days for whipworms, roundworms, hookworms
  • Child dosage: 1 teaspoonful single dose for threadworms
  • Child dosage: 1 teaspoonful twice daily for 3 days for whipworms, roundworms, hookworms
  • Reinfection may require a second dose after 2 weeks

Interaction

Preliminary evidence of cimetidine inhibiting Promex metabolism and potentially increasing plasma concentration

Contraindications

  • Contraindicated in patients with known hypersensitivity to Mebendazole
  • Contraindicated in patients with known hypersensitivity to any component of the formulation

Side Effects

  • Transient symptoms of abdominal pain and diarrhea
  • Hypersensitivity reactions: rash, urticaria, angioedema
  • Rare cases of convulsions
  • Neutropenia and agranulocytosis

Pregnancy & Lactation

  • Not recommended in pregnant women
  • Caution required when administered to nursing mothers due to potential excretion in human milk

Precautions & Warnings

  • Periodic assessment of organ system functions advisable during prolonged therapy
  • Patients should be informed of potential risk to the fetus in women taking Promex during pregnancy, especially during the first trimester
  • Cleanliness important to prevent reinfection and transmission of the infection

Use in Special Populations

  • The drug has not been extensively studied in children under two years
  • Relative benefit/risk should be considered in the treatment of children under two years

Overdose Effects

Accidental overdosage may cause gastrointestinal complaints lasting up to a few hours; vomiting and purging should be induced
