Description
- Pantoprazole is a proton pump inhibitor indicated for the treatment of peptic ulcer diseases, gastroesophageal reflux diseases, ulcer induced by non-steroidal anti-inflammatory drugs (NSAIDs), eradication of Helicobacter pylori, and Zollinger-Ellison Syndrome.

Pharmacology
- Proton pump inhibitor
- Suppresses gastric acid production
- Binds to H+/K+ATPase enzyme system
Dosage & Administration
- Oral administration
- Benign gastric ulcer 40 mg daily
- Gastro-esophageal reflux disease 20-40 mg daily
- Duodenal ulcer 40 mg daily
- Prophylaxis of NSAID-associated gastric or duodenal ulcer 20 mg daily
- Zollinger-Ellison Syndrome 80 mg once daily
- IV Injection for duodenal ulcer and gastric ulcer 40 mg once daily for 7-10 days
- IV Injection for prevention of rebleeding in peptic ulcer 80 mg followed by infusion
- IV Injection for prophylaxis of acid aspiration 80 mg every 12 h for 24 h
- IV Injection for long-term management of Zollinger-Ellison Syndrome 80 mg every 12 hours
Interaction
- No significant drug interactions observed in clinical studies
Contraindications
- Hypersensitivity to any components of the formulation
Side Effects
- Well tolerated in short-term and long-term treatment
- Common side effects: headache, diarrhea
- Rare side effects: abdominal pain, flatulence, rash, insomnia, hyperglycemia
Pregnancy & Lactation
- FDA Pregnancy Category B
- No adequate and well-controlled studies in pregnant women
- Excreted in human milk
Precautions & Warnings
- Tablet should not be split, chewed or crushed
- Long-term therapy may lead to malabsorption of cyanocobalamin (Vitamin B12)
- Increased risk of osteoporosis related disorders
Overdose Effects
- No known symptoms of overdosage in humans
- Not readily dialyzable
- Symptomatic and supportive management recommended

Reconstitution
- IV injection only
- Slow intravenous injection
- Storage at room temperature for a maximum 4 hours
- Intravenous infusion over approximately 15 minutes
- Storage at room temperature for a maximum 4 hours prior to further dilution
