Indications
- Palliative treatment of advanced carcinoma of the breast or endometrium
- Treatment of anorexia, cachexia, or weight loss in patients with AIDS & cancer
Pharmacology
- Synthetic, antineoplastic and progestational drug
- Mechanism of action involves inhibition of pituitary gonadotrophin production and decrease in estrogen secretion
- Antineoplastic action on breast carcinoma by modifying the action of steroid hormones and exerting direct cytotoxic effect on tumor cells
- Estimates of plasma levels and excretion in urine and feces
Dosage & Administration
- Tablet dosage for breast cancer and endometrial carcinoma
- Oral suspension dosage for anorexia, cachexia, or weight loss in patients with AIDS & cancer
Interaction
- Potential interactions with concomitant medications and food
- Possible fluid retention with estrogens and progestogens
Contraindications
- Hypersensitivity to Megestrol Acetate or any component
- Known or suspected pregnancy
Side Effects
- Weight gain
- Thromboembolic phenomena
- Glucocorticoid effects
- Other side effects like nausea, dyspnea, hyperglycemia, rash, etc.
Pregnancy & Lactation
- Pregnancy Category D
- Not recommended during the first four months of pregnancy
- Excretion in mother's milk
- Discontinuation of nursing during treatment
Precautions & Warnings
- Close surveillance for patients with recurrent or metastatic cancer
- Use with caution in patients with a history of thromboembolic disease
- Exacerbation of preexisting diabetes
Use in Special Populations
- Safety and effectiveness in pediatric patients have not been established
- Insufficient data for patients 65 years and older
Overdose Effects
- No serious unexpected side effects reported from high dosages
