Description
- Metformin tablets are used for the treatment of type 2 diabetes mellitus in adults and children over 10 years of age. It helps in managing blood glucose levels and improving insulin sensitivity.

Dosage Administration

Pharmacology
- Biguanide type oral antihyperglycemic drug
- Lowers both basal and postprandial plasma glucose
- Improves insulin sensitivity and peripheral glucose uptake
Interaction
- Potential interactions with drugs like Furosemide and Nifedipine
- Caution with cationic drugs eliminated by renal tubular secretion
- Certain drugs may lead to loss of glycemic control
Contraindications
- Hypersensitivity to the active substance or excipients
- Acute metabolic acidosis, severe renal failure, acute conditions altering renal function
- Acute or chronic diseases causing tissue hypoxia
- Not advised in conditions causing dehydration or in patients with serious infections or trauma
Side Effects
- Hemolytic anemia

- Decrease of vitamin B12 absorption
- Peripheral neuropathy in patients with vitamin B12 deficiency
- Taste disturbance
- Encephalopathy
- Gastrointestinal disorders like nausea, vomiting, diarrhea, abdominal pain
- Liver function test abnormalities or hepatitis
- Skin reactions like erythema, pruritus, urticaria
Pregnancy and Lactation
- Uncontrolled diabetes during pregnancy increases risk of congenital abnormalities and perinatal mortality
- Not recommended during pregnancy but insulin can be used
- Excreted into human breast milk
- Breastfeeding not recommended during treatment
Precautions and Warnings
- Substantially excreted by the kidney
- Increases risk of lactic acidosis and hypoglycemia in combination with insulin
- Regular monitoring of renal function advised
- Caution with dehydration, serious infections, trauma
- Annual estimation of Vitamin B-12 levels recommended
- Stabilization of diabetic patients with Metformin and insulin should be in hospital
Use in Special Populations
- Dosage adjustment based on renal function in elderly subjects
- Confirmation of type 2 diabetes mellitus before treatment initiation in children
- No effect on growth and puberty detected in controlled clinical studies for a year
- Renal function assessment and caution in elderly subjects
- Contraindicated in patients with GFR less than 30 mL/min
Overdose Effects
- Lactic acidosis observed in overdose situations
- Hemodialysis recommended for removal of lactate and Metfomin
