Indications
- Peptic ulcer diseases
- Gastroesophageal reflux diseases
- Ulcer induced by non-steroidal anti-inflammatory drugs (NSAIDs)
- Eradication of Helicobacter pylori (in combination with antibiotics)
- Zollinger-Ellison Syndrome
Pharmacology
- Proton pump inhibitor
- Suppresses gastric acid production
- Covalently binds to H+/K+ ATPase enzyme system
- Inhibits both basal and stimulated gastric acid secretion
Dosage and administration
- Dosage for benign gastric ulcer: 40 mg daily in the morning for 4 weeks, continued for further 4 weeks if not fully healed
- Dosage for gastro-esophageal reflux disease: 20-40 mg daily in the morning for 4 weeks, continued for further 4 weeks if not fully healed
- Dosage for duodenal ulcer: 40 mg daily in the morning for 2 weeks, continued for further 2 weeks if not fully healed
- Dosage for duodenal ulcer associated with Helicobacter pylori: 40 mg twice daily in combination with antibiotics for one week
- Dosage for prophylaxis of NSAID-associated gastric or duodenal ulcer: 20 mg daily
- Dosage for Zollinger-Ellison Syndrome: Initially 80 mg once daily adjusted according to response, with a maximum of 40 mg daily for elderly patients
Interaction
- No significant drug interactions observed in clinical studies
Contraindications
- Not for use in patients with known hypersensitivity to any of the components
Side effects
- Common side effects: Headache, diarrhea
- Rare side effects: Abdominal pain, flatulence, rash, insomnia, hyperglycemia
Pregnancy and lactation
- Category B for pregnancy
- Excreted in human milk
- Use during pregnancy only if clearly needed
Precautions and warnings
- Tablet should not be split, chewed or crushed
- Long-term therapy may lead to malabsorption of cyanocobalamin (Vitamin B12)
- Increased risk of osteoporosis related disorders
Overdose effects
- No known symptoms of overdosage in humans
- Not readily dialyzable
- No specific therapy recommended

Reconstitution
- IV injection should be given as a slow intravenous injection
- Reconstituted solution may be stored at room temperature for a maximum of 4 hours
- IV infusion should be given over a period of approximately 15 minutes
- Reconstituted solution may be stored at room temperature for a maximum of 4 hours prior to further dilution
