Pharmacology
- Mediated through the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Increase the activity of GABA
- Produce a sedating effect
- Relax skeletal muscles
- Induce sleep, anesthesia, and amnesia
- Bind to the benzodiazepine site on GABA-A receptors
- Potentiate the effects of GABA by increasing the frequency of chloride channel opening
Dosage & Administration
- Oral dosage for adults: 7.5-15 mg daily
- Oral dosage in elderly and debilitated patients: 7.5 mg
- Premedication dosage: 15 mg 30-60 minutes before the procedure
- Intravenous dosage for endoscopic or cardiovascular procedures in healthy adults: approximately 2.5 mg
- Intravenous dosage for induction of anesthesia: 10-15 mg
- Intramuscular dosage for adults: 0.07-0.1 mg/kg body weight
- Intramuscular dosage for children: 0.15-0.20 mg/kg
- Intramuscular dosage for elderly and debilitated patients: 0.025-0.05 mg/kg
- Rectal administration dosage in children for preoperative sedation: 0.35-0.45 mg/kg 20-30 min before induction of general anesthesia

Contraindications
- Severe respiratory insufficiency
