Indications
- Peptic ulcer diseases
- Gastroesophageal reflux diseases
- Ulcer induced by non-steroidal anti-inflammatory drugs (NSAIDs)
- Eradication of Helicobacter pylori (in combination with antibiotics)
- Zollinger-Ellison Syndrome
Pharmacology
- Proton pump inhibitor
- Final step in gastric acid production suppression
- Covalent binding to H+/K+ATPase enzyme system
- Inhibition of both basal and stimulated gastric acid secretion
Dosage & Administration
- 40 mg daily dosage for benign gastric ulcer
- 20-40 mg daily dosage for gastro-esophageal reflux disease
- 40 mg daily dosage for duodenal ulcer
- 40 mg twice daily dosage for duodenal ulcer associated with Helicobacter pylori
- 20 mg daily dosage for prophylaxis of NSAID-associated gastric or duodenal ulcer
- Initial 80 mg daily dosage for Zollinger-Ellison Syndrome
Interaction
- No significant drug interactions observed in clinical studies
Contraindications
- Known hypersensitivity to any of the components of the formulation
Side Effects
- Well tolerated in both short-term and long-term treatment
- Common: headache, diarrhea
- Rare: abdominal pain, flatulence, rash, insomnia, hyperglycemia
Pregnancy & Lactation
- US FDA Pregnancy Category: B
- No adequate and well-controlled studies in pregnant women
- Excreted in human milk
- Use during pregnancy only if clearly needed, consider benefit to the mother when nursing
Precautions & Warnings
- Tablet should not be split, chewed or crushed
- Long-term therapy may lead to malabsorption of cyanocobalamin (Vitamin B12) and increased risk of osteoporosis-related disorders
Overdose Effects
- No known symptoms of overdosage in humans
- Highly protein bound, not readily dialyzable
- Symptomatic and supportive management recommended
Reconstitution
- IV administration only
- Slow intravenous injection
- Reconstituted solution may be stored at room temperature for a maximum of 4 hours
- Intravenous infusion over a period of approximately 15 minutes
