Indications

  • Peptic ulcer diseases
  • Gastroesophageal reflux diseases
  • Ulcer induced by non-steroidal anti-inflammatory drugs (NSAIDs)
  • Eradication of Helicobacter pylori (in combination with antibiotics)
  • Zollinger-Ellison Syndrome

Pharmacology

  • Proton pump inhibitor
  • Final step in gastric acid production suppression
  • Covalent binding to H+/K+ATPase enzyme system
  • Inhibition of both basal and stimulated gastric acid secretion

Dosage & Administration

  • 40 mg daily dosage for benign gastric ulcer
  • 20-40 mg daily dosage for gastro-esophageal reflux disease
  • 40 mg daily dosage for duodenal ulcer
  • 40 mg twice daily dosage for duodenal ulcer associated with Helicobacter pylori
  • 20 mg daily dosage for prophylaxis of NSAID-associated gastric or duodenal ulcer
  • Initial 80 mg daily dosage for Zollinger-Ellison Syndrome

Interaction

  • No significant drug interactions observed in clinical studies

Contraindications

  • Known hypersensitivity to any of the components of the formulation

Side Effects

  • Well tolerated in both short-term and long-term treatment
  • Common: headache, diarrhea
  • Rare: abdominal pain, flatulence, rash, insomnia, hyperglycemia

Pregnancy & Lactation

  • US FDA Pregnancy Category: B
  • No adequate and well-controlled studies in pregnant women
  • Excreted in human milk
  • Use during pregnancy only if clearly needed, consider benefit to the mother when nursing

Precautions & Warnings

  • Tablet should not be split, chewed or crushed
  • Long-term therapy may lead to malabsorption of cyanocobalamin (Vitamin B12) and increased risk of osteoporosis-related disorders

Overdose Effects

  • No known symptoms of overdosage in humans
  • Highly protein bound, not readily dialyzable
  • Symptomatic and supportive management recommended

Reconstitution

  • IV administration only
  • Slow intravenous injection
  • Reconstituted solution may be stored at room temperature for a maximum of 4 hours
  • Intravenous infusion over a period of approximately 15 minutes
