Indications
- Short-term treatment of insomnia
- Sedation in premedication before surgical or diagnostic procedures
Description
Sleep-inducing agent characterized by rapid onset and short duration of action; exerts anxiolytic, anticonvulsant, and muscle-relaxant effects
Pharmacology
Mediated through the inhibitory neurotransmitter gamma-aminobutyric acid (GABA); increases GABA activity, producing a sedating effect, relaxing skeletal muscles, inducing sleep, anesthesia, and amnesia
Dosage & Administration
- Oral dosage for adults: 7.5-15 mg daily
- In elderly and debilitated patients: The recommended dose is 7.5 mg
- In premedication: 15 mg of Midazolam should be given 30-60 minutes before the procedure
- Intravenous administration for endoscopic or cardiovascular procedures: Initial dose is approximately 2.5 mg; reduced to 1-1.5 mg for severe illness and elderly patients
- Induction of anesthesia: 10-15 mg
- Intramuscular administration for adults: 0.07-0.1 mg/kg body weight; usual dose is about 5 mg
- Rectal administration in children for preoperative sedation: 0.35-0.45 mg/kg 20-30 min. before induction of general anesthesia
Interaction
Enhances the central sedative effect of neuroleptics, tranquillizers, antidepressants, sleep-inducing drugs, analgesics, anaesthetics, antipsychotics, anxiolytics, antiepileptic drugs, and sedative antihistamines
Contraindications
Severe respiratory insufficiency, severe hepatic insufficiency, myasthenia gravis, sleep apnea syndrome, known hypersensitivity to benzodiazepines or any component of the product

Pregnancy & Lactation
Should be avoided during pregnancy unless no safer alternative; passes into breast milk, should not be administered to breastfeeding mothers
Precautions & Warnings
IV administration should be very slow
Overdose Effects
Extreme overdosage may lead to coma, areflexia, cardiorespiratory depression, and apnea; controlled with benzodiazepine antagonist flumazenil
