Indications

  • Short-term treatment of insomnia

Description

A sleep-inducing agent with rapid onset and short duration of action, exerting an anxiolytic, anticonvulsant, and muscle-relaxant effect

Pharmacology

Actions are mediated through the inhibitory neurotransmitter gamma-aminobutyric acid (GABA), inducing sedating effect, relaxing skeletal muscles, and inducing sleep, anesthesia, and amnesia

Dosage & Administration

  • Oral dosage for adults: 7.5-15 mg daily
  • Intravenous administration for endoscopic or cardiovascular procedures: initial dose is approximately 2.5 mg in healthy adults

Interaction

Can enhance the central sedative effect of neuroleptics, tranquillizers, antidepressants, sleep-inducing drugs, analgesics, anaesthetics, antipsychotics, anxiolytics, antiepileptic drugs, and sedative antihistamines

Contraindications

Must not be given to patients with severe respiratory or hepatic insufficiency, myasthenia gravis, sleep apnea syndrome, or known hypersensitivity to benzodiazepines or any component of the product

Side Effects

  • Drowsiness, confusion, fatigue, headache, and muscle weakness may occur at the start of therapy
  • Fluctuations in vital signs including respiratory depression and variations in blood pressure and pulse rate have been noted following parenteral administration

Pregnancy & Lactation

Should be avoided during pregnancy unless no safer alternative, and should not be administered to breastfeeding mothers

Precautions & Warnings

Dormitol IV should be administered very slowly

Overdose Effects

Extreme overdosage may lead to coma, cardiorespiratory depression, and apnea, and can be controlled with benzodiazepine antagonist flumazenil
