Indications
- Prophylaxis and chronic treatment of asthma
- Acute prevention of Exercise-Induced Bronchoconstriction (EIB)
- Relief of symptoms of Allergic Rhinitis (AR): Seasonal & Perennial Allergic Rhinitis
Pharmacology
- Selective and orally active leukotriene receptor antagonist
- Inhibition of the cysteinyl leukotriene receptor (CysLT1)
- Correlation with the pathophysiology of asthma & allergic rhinitis
- Inhibition of airway edema, smooth muscle contraction, and altered cellular activity
Dosage & Administration
- 10 mg tablet once daily for adults and adolescents 15 years of age and older
- 5 mg tablet once daily for pediatric patients 6 to 14 years of age
- 4 mg tablet or oral granules once daily for pediatric patients 2 years to 5 years of age
- Use in pediatric patients aged 6 months to 14 years
- No dosage adjustment in hepatic or renal insufficiency
- Similar pharmacokinetic profile and oral bioavailability in elderly and younger adults
Interaction
- No increase in adverse reactions when administered with other therapies for asthma
- No clinically important effects on the pharmacokinetics of certain drugs
- Used concomitantly with a wide range of commonly prescribed drugs without evidence of clinical adverse interactions
- Potential AUC decrease with phenobarbital induction of hepatic metabolism
Contraindications
- Hypersensitivity to any component of the product
Side Effects
- Common: Diarrhoea, fever, headache, nausea, vomiting, skin reactions, upper respiratory tract infection
- Uncommon: Akathisia, anxiety, arthralgia, depression, dizziness, dry mouth, malaise, muscle complaints, seizure
- Rare: Angioedema, disorientation, hallucination, memory loss, palpitations, suicidal tendencies, tremor
Pregnancy & Lactation
- Crosses the placenta following oral dosing in rats and rabbits
- Caution advised during pregnancy and lactation
Precautions & Warnings
- Not for use in the reversal of bronchospasm in acute asthma attacks
- Appropriate rescue medication should be available
- Can be continued during acute exacerbations of asthma
- Should not be abruptly substituted for inhaled or oral corticosteroids
- Not for monotherapy in the treatment and management of exercise-induced bronchospasm
- Continued avoidance of aspirin or non-steroidal anti-inflammatory agents for patients with known aspirin sensitivity
Overdose Effects
- Majority of reports had no adverse experiences
- Usual supportive measures recommended in the event of overdose
