Indications
- Treatment of active duodenal ulcer
- Treatment of active benign gastric ulcer

Pharmacology
- Suppresses gastric acid secretion
- Inhibits the gastric H+/K+-ATPase at the secretory surface of the gastric parietal cell
- Characterized as a gastric proton-pump inhibitor
Dosage
- Recommended dose for active duodenal ulcer and active benign gastric ulcer: 20 mg once daily in the morning
- Recommended dose for erosive or ulcerative gastro-esophageal reflux disease (GERD): 20 mg once daily for 4-8 weeks
- Recommended dose for gastro-esophageal reflux disease long-term management: 20 mg or 10 mg once daily
- Recommended dose for treatment of symptomatic GERD: 10 mg once daily
- Recommended dose for treatment of Zollinger-Ellison Syndrome: 60 mg once daily, may titrate upwards to 120 mg/day
- Recommended dose for eradication of H. pylori: Rabeprazole sodium 20 mg twice daily
Administration
- Tablets should be taken in the morning before eating
- Tablets should not be chewed or crushed, but should be swallowed whole
Interaction
- Potential interaction with compounds whose absorption is pH dependent
- Potential interaction with ketoconazole or itraconazole leading to decreased antifungal plasma levels
Contraindications
- Hypersensitivity to the active substance or any of the excipients
- Contra-indicated in pregnancy and during breastfeeding

Precautions & Warnings
- Symptomatic response does not preclude the presence of gastric or oesophageal malignancy
- Regular surveillance for long-term treatment patients
- Risk of hip, wrist, and spine fracture with long-term high-dose use
- Risk of cross-hypersensitivity reactions with other proton pump inhibitor or substituted benzimidazoles
- Caution in patients with mild to moderate hepatic impairment
- Hypomagnesaemia reported in patients treated for at least three months
- Reduction in vitamin B12 absorption
Use in Special Populations
- No dosage adjustment necessary for patients with renal or hepatic impairment
- Not recommended for use in children
Overdose Effects
- Effects are generally minimal and reversible without further medical intervention
- No specific antidote known
- Not dialysable due to extensive protein binding
