Dosage
- 20 mg to be taken once daily in the morning for active duodenal ulcer and active benign gastric ulcer
- 20 mg to be taken once daily for four to eight weeks for erosive or ulcerative gastro-esophageal reflux disease (GERD)
- Maintenance dose of 20 mg or 10 mg once daily for GERD maintenance
- 10 mg once daily for symptomatic treatment of moderate to very severe GERD in patients without oesophagitis
- 5 mg once daily for 12 weeks for treatment of GERD in pediatric patients 1 to 11 years of age (Less than 15 kg)
- 10 mg once daily for 12 weeks for treatment of GERD in pediatric patients 1 to 11 years of age (15 kg or more)
- 60 mg once a day for Zollinger-Ellison Syndrome
- Rabeprazole sodium 20 mg twice daily, clarithromycin 500 mg twice daily, and amoxicillin 1g twice daily for eradication of H. pylori
Administration
- To be taken in the morning, before eating; should be swallowed whole
Interaction
- Possible decrease in antifungal plasma levels when co-administered with ketoconazole or itraconazole
- No interaction with liquid antacids observed
- Interaction with atazanavir, as it is pH-dependent
Contraindications
- Hypersensitivity to the active substance or excipients
- Contra-indicated in pregnancy and during breastfeeding
Side effects
- Well-tolerated in short-term and long-term studies
- May cause headache, diarrhea, abdominal pain, vomiting, constipation, dry mouth, changes in appetite, muscle pain, drowsiness, dizziness
Pregnancy lactation
- US FDA pregnancy category 'C'
- No adequate and well-controlled studies in pregnant women
- Likely to be excreted in human milk
Precautions warnings
- Symptomatic response to therapy does not preclude the presence of malignancy
- Patients on long-term treatment should be under regular surveillance
- May modestly increase the risk of hip, wrist, and spine fracture, particularly in the elderly or with other risk factors
- A risk of cross-hypersensitivity reactions with other proton pump inhibitor or substituted benzimidazoles
- Hepatic enzyme abnormalities seen in clinical trials
- Co-administration with atazanavir not recommended
- Possible increase in the risk of gastrointestinal infections such as Salmonella, Campylobacter, and Clostridium difficile with proton pump inhibitors
- Risk of hypomagnesaemia and influence on vitamin B12 absorption
Renal hepatic impairment
- No dosage adjustment necessary for patients with renal or hepatic impairment

- Effects are generally minimal and reversible without further medical intervention
- No specific antidote known
