Pharmacology
- Blocks COX-1 and COX-2 enzymes
- Decreased prostaglandin synthesis
- COX-1 enzyme mediates gastrointestinal upset and renal toxicities
- COX-2 enzyme mediates antipyretic, analgesic and anti-inflammatory properties
Dosage & Administration
- Should be taken with water after meals
- Dosage for adults with rheumatoid arthritis/osteoarthritis/ankylosing spondylitis
- Dosage for acute gout
- Dosage for dysmenorrhoea
- Dosage for other indications like analgesia and acute musculoskeletal disorders

Interaction
- Diminishes the antihypertensive effect of ACE inhibitors
- Delays the absorption of Naxin SR by antacids & sucralfate
- Increases adverse effects with aspirin
- Reduces the natriuretic effect of diuretics
- Enhances the toxicity of methotrexate
- Increases the risk of GI bleeding with warfarin and SSRIs
Contraindications
- Known hypersensitivity to Naproxen
- Patients who have experienced asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- Perioperative pain in the setting of coronary artery bypass graft (CABG) surgery
Side Effects
- Gastrointestinal side effects: Heartburn, abdominal pain, nausea, diarrhea, dyspepsia
- Central Nervous System side effects: Headache, vertigo, drowsiness
- Dermatological side effects: Pruritus (itching), purpura
- Cardiovascular side effects: Edema, palpitation
- Other side effects: Visual disturbances, hearing disturbances
Pregnancy & Lactation
- Adverse effects on the fetus
- Causes a delay in parturition in animals
- Associated with premature closure of the ductus arteriosus
- Excreted in small amounts in the breast milk
Precautions & Warnings
- Risk of renal papillary necrosis and abnormal renal pathology
- Reports of acute interstitial nephritis, hematuria, proteinuria
- Reports of moderate to severe jaundice attributed to Naxin SR
- Caution for patients with significantly impaired renal function
- Monitoring of serum creatinine and/or creatinine clearance advised
- Caution for patients with creatinine clearance of less than 20ml/min
