Indications
- Osteoarthritis
- Rheumatoid arthritis
- Ankylosing spondylitis
- Decrease the risk of developing gastric ulcers
Pharmacology
- Immediate release Esomeprazole Magnesium layer
- Enteric-coated Naproxen core
- Proton pump inhibitor
- Inhibits gastric acid secretion
- Suppresses gastric acid secretion
- Inhibits the prostaglandin synthesis
Dosage
- Adult dosage: One tablet twice daily
- Juvenile Idiopathic Arthritis dosage based on weight
Administration
- Take at least 30 minutes before meals
- Do not split, chew, crush or dissolve the tablet
Interaction
- Concomitant use of NSAIDs may reduce the antihypertensive effect of ACE inhibitors, diuretics & beta-blockers
- Concomitant use with warfarin may increase the risk of bleeding
- Inhibition of gastric acid secretion may interfere with the absorption of certain drugs
- Administration with high-fat food affects absorption
Contraindications
- Known hypersensitivity to any component of the tablet
- History of allergic-type reactions after taking aspirin or other NSAIDs
- Use during the peri-operative period in the setting of coronary artery bypass graft surgery
Side Effects
- Gastrointestinal side effects
- Decreased incidence of gastric ulcers and NSAID associated upper gastrointestinal adverse events
- Increased risk of arterial thrombotic events
- Reported adverse events include oedema, hypertension, and cardiac failure
Pregnancy & Lactation
- Not recommended during the third trimester of pregnancy
- Should not be used during breastfeeding
- May impair female fertility
Precautions & Warnings
- Avoid use with other NSAIDs
- Minimize undesirable effects by using the lowest effective dose for the shortest duration
- Use with caution in older people and patients with specific conditions
Use in Special Populations
- Elderly patients may require dosage adjustment
- Not recommended for use in patients with moderate to severe renal impairment
- Monitor patients with hepatic insufficiency closely
Overdose Effects
- Symptoms may include lethargy, dizziness, epigastric pain, and gastrointestinal bleeding
- Patients should be managed by symptomatic and supportive care
- No specific antidote is known
