Indications
- Relief from chronic heartburn symptoms and other symptoms associated with GERD
- Healing of erosive esophagitis
- Maintenance of healing of erosive esophagitis
- Eradication of Helicobacter pylori infection in patients with duodenal ulcer disease
- Zollinger-Ellison Syndrome
- Treatment of Acid related Dyspepsia
- Treatment of Duodenal & Gastric ulcer
Pharmacology
- Proton pump inhibitor that suppresses gastric acid secretion
- Absorption peak plasma levels occur at approximately 1.5 hours after oral administration
- Metabolized extensively in the liver by the cytochrome P450 (CYP) enzyme system
- Plasma elimination half-life is approximately 1-1.5 hours
- Distribution: 97% bound to plasma proteins
Dosage
- Oral dosage form for healing of erosive esophagitis: 20 mg or 40 mg once daily for 4-8 weeks
- IV injection or IV infusion for GERD with erosive esophagitis: 20 mg or 40 mg once daily for adults
- Pediatric dosage: 10 mg once daily for patients with body weight <55 kg
- Dosage for risk reduction of rebleeding of Gastric or Duodenal Ulcers following therapeutic endoscopy: 80 mg for adults
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Oral suspension should be taken into a small glass containing 15 ml of water and stirred well
- IV Injection must be administered intravenously over a period of at least 3 minutes
Interaction
- Extensively metabolized in the liver by CYP2C19 and CYP3A4
- No clinically significant interactions with phenytoin, warfarin, quinidine, clarithromycin or amoxicillin
Contraindications
- Contraindicated in-patient with known hypersensitivity to any of the formulation
Side Effects
- Most frequent adverse events include headache, diarrhea, nausea, flatulence, abdominal pain, constipation, and dry mouth
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Animal studies have revealed no teratogenic effects
- Breastfeeding should be discontinued if the use of esomeprazole is considered essential
Precautions & Warnings
- Symptomatic response to therapy with esomeprazole does not preclude the presence of gastric malignancy
- Capsules should be taken at least one hour before meals
Use in Special Populations
- Safety and effectiveness in pediatric patients have not been established
- No overall differences in safety and efficacy between the elderly and younger individuals
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
Overdose Effects
- Signs of acute toxicity include reduced motor activity, changes in respiratory frequency, tremor, and intermittent clonic convulsions

- Store at a temperature not exceeding 30°C in a dry place
