Medicine Details

Indications

  • Relief of signs and symptoms of osteoarthritis
  • Relief of signs and symptoms of rheumatoid arthritis
  • Relief of signs and symptoms of ankylosing spondylitis
  • Decrease the risk of developing gastric ulcers in patients at risk of developing NSAID-associated gastric ulcers

Pharmacology

  • Consists of an immediate release esomeprazole magnesium layer and an enteric-coated naproxen core
  • Naproxen is a NSAID with analgesic and antipyretic properties
  • Esomeprazole is a proton pump inhibitor that suppresses gastric acid secretion

Dosage

  • Use the lowest effective dose for the shortest duration consistent with individual patient treatment goals
  • Different dosage options for adults and juvenile idiopathic arthritis in adolescent patients
  • Tablet to be taken at least 30 minutes before meals

Administration

  • Tablet is not to be split, chewed, crushed or dissolved

Interaction

  • Concomitant use of NSAIDs may reduce the antihypertensive effect of ACE inhibitors, diuretics, and beta-blockers
  • Concomitant use of this tablet and warfarin may result in an increased risk of a bleeding complication
  • Esomeprazole may interfere with the absorption of certain drugs

Contraindications

  • Known hypersensitivity to any component of this tablet or substituted benzimidazoles
  • History of asthmay urticaria or other allergic-type reactions after taking aspirin or other NSAIDs
  • Use during the peri-operative period in the setting of coronary artery bypass graft (CABG) surgery

Side Effects

  • Decrease the incidence of gastrointestinal side effects from naproxen
  • Naproxen has been associated with risk of arterial thrombotic events
  • Gastrointestinal adverse events may occur

Pregnancy & Lactation

  • Caution in use during pregnancy and contraindicated during the third trimester
  • Naproxen is excreted in low quantities in human milk
  • Use of NSAIDs may impair female fertility

Precautions & Warnings

  • Combination with NSAIDs should be avoided
  • Risk-factors to develop NSAID related gastro-intestinal complications
  • Caution advised for older people and patients with gastrointestinal disease

Use in Special Populations

  • Caution required in elderly patients and those with moderate to severe renal impairment
  • Hepatic insufficiency should be monitored closely

Overdose Effects

  • Symptoms of overdose and their management for both naproxen and esomeprazole
