Pharmacology
- Inhibitor of gastric acid secretion

- Inhibition of secretion lasts up to 72 hours

Interaction
- Decreased intragastric acidity affecting the absorption of ketoconazole
- Metabolized in the liver through cytochrome P450
- Potential interaction with drugs like ketoconazole, diazepam, phenytoin, warfarin, clarithromycin, amoxicillin, metronidazole
- No evidence of interaction with phenacetin, theophylline, caffeine, propranolol, metoprolol, cyclosporin, lidocaine, quinidine, estradiol, antacids, piroxicam, diclofenac, naproxen
Contraindications
- Known hypersensitivity
- Suspicion of gastric ulcer without excluding the possibility of malignancy

- Photosensitivity, bullous eruption, erythema multiforme, angioedema, alopecia
- Diarrhea, headache, constipation, nausea/vomiting, flatulence, abdominal pain
- Dry mouth, stomatitis, candidiasis
- Paraesthesia, dizziness, lightheadedness, faintness
- Somnolence, insomnia, vertigo
- Reversible mental confusion, agitation, depression, hallucinations
- Arthritic and myalgic symptoms
- Blurred vision, taste disturbance, peripheral edema, increased sweating, gynaecomastia, hematological disorders, anaphylactic shock, malaise, fever, bronchospasm, hepatic disorders
Pregnancy & Lactation
- No adverse effects on pregnancy or the health of the fetus/newborn child
- Can be used during pregnancy

Precautions & Warnings
- Avoid concomitant use with clopidogrel
- Possible association with increased risk for osteoporosis-related fractures
