Indications
- Prevention of nausea and vomiting associated with initial and repeat courses of emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Mode of Action
- Selective blocking agent of the serotonin 5-HT3 receptor type
- Blocks initiation of vomiting reflex by activating vagal afferents via 5HT3 receptors
- Antagonism of 5HT3 receptors on neurons located in the peripheral and central nervous system
Dosage
- Adults, Pediatric patients (6 months to 18 years) - 8 mg or 4 mg tablet/orodispersible tablet for chemotherapy-induced nausea and vomiting
- Adults - 8 mg tablet/orodispersible tablet for radiotherapy-induced nausea and vomiting
- Adults - 8 mg or 4 mg orodispersible tablet for postoperative nausea and vomiting
- Pediatrics (>40 kg) - Injection for postoperative nausea and vomiting
- Pediatrics (40 kg) - Injection for postoperative nausea and vomiting
- Various dosage for different types of cancer chemotherapy and radiotherapy

Administration
- Oral soluble film designed to be applied on top of the tongue and dissolve within 20 seconds

Interaction
Leofran does not induce or inhibit the cytochrome P-450 drug-metabolizing enzyme system of the liver. Interactions with other drugs may affect the clearance and half-life of Leofran.
Contraindications
Contraindicated in patients known to have hypersensitivity to the drug or any of its components. Concomitant use of apomorphine.
Side Effects
- Frequently reported adverse events include headache, constipation, and diarrhea
- Rash, sensation of flushing or warmth, hiccups, and liver enzyme abnormalities reported
- Rare cases of anaphylaxis, brochospasm, tachycardia, angina, hypokalemia, and shortness of breath reported
Pregnancy & Lactation
- No evidence of impaired fertility or harm to fetus in animal studies
- Caution advised when administered to nursing women
Precautions & Warnings
- Hypersensitivity reactions reported in patients with hypersensitivity to other selective 5-HT3 receptor antagonists
- Not a drug that stimulates gastric or intestinal peristalsis
- Caution required in patients following abdominal surgery or with chemotherapy-induced nausea and vomiting
Use in Special Populations
- Dosage adjustment for patients with impaired renal function
- Dosage adjustment for patients with impaired hepatic function
- Limited information available about dosage in pediatric patients 4 years of age or younger
