Indications
- Prevention of nausea and vomiting associated with initial and repeat courses of emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Dosage
- Available in 8 mg orodispersible tablet
- Available in 4 mg orodispersible tablet
- Administered orally, intravenously or as an oral solution based on patient's age and condition
Mode of Action
- Blocks the initiation of vomiting reflex by activating vagal afferents via 5HT3 receptors
- Antagonizes 5HT3 receptors on neurons located in the peripheral and central nervous system
Administration
- Oral soluble film that dissolves within 20 seconds when placed on the tongue
- Film does not require water to aid dissolution or swallowing
- Can be swallowed with or without liquid
Interaction
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
- Dosage adjustment may be required for patients on drugs that induce or inhibit these enzymes
Contraindications
- Hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events include headache, constipation, and diarrhea
- Rare cases of anaphylaxis, tachycardia, and shortness of breath reported
Pregnancy & Lactation
- No evidence of impaired fertility or harm to the fetus in animal studies
- Excreted in the breast milk of rats, caution advised when administered to nursing women
Precautions & Warnings
- Should not be used instead of nasogastric suction
- May mask a progressive ileus and/or gastric distension in specific patient groups
Use in Special Populations
- Dosage adjustments recommended for patients with impaired hepatic function
- Limited information available for pediatric patients 4 years of age or younger
