Indications
- Prevention of nausea and vomiting associated with initial and repeat courses of emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Description
Orally dissolving film designed to be applied on top of the tongue where it will dissolve within 20 seconds and then swallowed with saliva. Does not require water to aid dissolution or swallowing.
Active Ingredient
Vomidyl base, the racemic form of Vomidyl, and a selective blocking agent of the serotonin 5-HT3 receptor type
Mode of Action
Blocks 5HT3 receptors on neurons located both in the peripheral and central nervous system. Mechanisms of action in post-operative nausea and vomiting are not known.
Dosage
- Dosage varies for different conditions and age groups. Includes tablets, orodispersible tablets, and oral solution. Administration specifics are provided.
Administration Steps
- Step 1: Tear the pouch carefully along the edge tear mark
- Step 2: Place the film on top of the tongue. It will dissolve within 20 seconds
- Step 3: Do not chew or swallow the film whole
- Step 4: Swallow after the film dissolves. Can be swallowed with or without liquid
- Step 5: Wash hands after taking the film
Interaction
Does not appear to induce or inhibit the cytochrome P-450 drug-metabolizing enzyme system of the liver. Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes.
Contraindications
- Hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events include headache, constipation, and diarrhea. Other reported effects include rash, sensation of flushing or warmth, hiccups, liver enzyme abnormalities, anaphylaxis, brochospasm, tachycardia, angina, hypokalemia, shortness of breath.
Pregnancy & Lactation
Carcinogenic effects were not seen in 2-year studies in rats and mice. Not mutagenic in standard tests. Caution advised when administered to nursing women.
Precautions & Warnings
- Hypersensitivity reactions have been reported in patients who have exhibited hypersensitivity to other selective 5-HT3 receptor antagonists
- Vomidyl should not be used instead of nasogastric suction
- Use in patients following abdominal surgery or with chemotherapy-induced nausea and vomiting may mask a progressive ileus and/or gastric distension.
Use in Special Populations
- Dosage adjustment not needed for patients with impaired renal function
- Dosage adjustment needed for patients with severe hepatic impairment
- Little information is available about dosage in pediatric patients 4 years of age or younger
