Indications
- Adjuvant treatment of stage III colon cancer in patients who have undergone complete resection of the primary tumor
- Treatment of advanced colorectal cancer
Pharmacology
- Undergoes nonenzymatic conversion in physiologic solutions
- Forms transient reactive species that covalently bind with macromolecules
- Forms Pt-DNA crosslinks inhibiting DNA replication and transcription
- Cell-cycle nonspecific cytotoxicity
Dosage & Administration
- Administer every 2 weeks
- For advanced disease, treatment is recommended until disease progression or unacceptable toxicity
- For adjuvant use, treatment is recommended for a total of 6 months (12 cycles)
- Specific dosages for Day 1 and Day 2 administration
Interaction
- No specific cytochrome P-450-based drug interaction studies conducted
- Increases of 5-fluorouracil plasma concentrations by approximately 20% with certain dosages
- Platinum-containing species elimination primarily through the kidney
Contraindications
- Contraindicated in patients with known allergy to oxaliplatin or other platinum compounds
