Indications

  • Adjuvant treatment of stage III colon cancer in patients who have undergone complete resection of the primary tumor
  • Treatment of advanced colorectal cancer

Pharmacology

  • Undergoes nonenzymatic conversion in physiologic solutions
  • Forms transient reactive species that covalently bind with macromolecules
  • Forms Pt-DNA crosslinks inhibiting DNA replication and transcription
  • Cell-cycle nonspecific cytotoxicity

Dosage & Administration

  • Administer every 2 weeks
  • For advanced disease, treatment is recommended until disease progression or unacceptable toxicity
  • For adjuvant use, treatment is recommended for a total of 6 months (12 cycles)
  • Specific dosages for Day 1 and Day 2 administration

Interaction

  • No specific cytochrome P-450-based drug interaction studies conducted
  • Increases of 5-fluorouracil plasma concentrations by approximately 20% with certain dosages
  • Platinum-containing species elimination primarily through the kidney

Contraindications

  • Contraindicated in patients with known allergy to oxaliplatin or other platinum compounds
