Indications
- Induction of labor
- Stimulation of uterine contractions
- Post delivery period for prevention or treatment of secondary haemorrhage and adherent placenta
- Stimulate lactation under advice of registered Gynaecologist/Physician
- Treatment of incomplete abortion to obtain more rapid expulsion
Description
- Synthetically prepared Inducin
- Identical to natural occurring hormone from posterior pituitary gland
- Causes contractions of uterus
- Impedes uterine blood flow
- Increases amplitude and duration of uterine contractions leading to dilation and effacement of the cervix
Pharmacology
- Uterine motility dependent on formation of contractile protein actomyosin
- Oxytocin promotes contractions by increasing intracellular Ca2+
- Oxytocin has specific receptors in muscle lining of uterus
Dosage & Administration
- Oxytocin given by intravenous infusion
- Initial administration at 1 mU per minute
- Dosage doubled every 30-45 minutes until uterus threshold attained
- Intramuscular administration may lead to irregular response
- Intramuscular dosage should not exceed 0.5-1 IU per injection
Interaction
- Severe hypertension reported when given following vasoconstrictor with caudal-block anesthesia
- Cyclopropane anesthesia may modify cardiovascular effects
- Maternal sinus bradycardia with abnormal atrioventricular rhythms noted when used concomitantly with cyclopropane anesthesia
Contraindications
- Cephalo-pelvic disproportion
- Abnormal position of the child or skull
- Overdistention of inferior segment of the uterus
- Severe toxaemia
- Hypertonic uterine dysfunction
- Foetal distress where delivery is not imminent
- Cord presentation and prolapse
- Total placenta previa
- Vasa praevia
Side Effects
- Hypersensitivity may result in uterine hypertonicity, spasm, or rupture
- Possibility of increased blood loss and afbrinigenemia
- Severe water intoxication with convulsions and coma
- May cause nausea, vomiting, haemorrhage, cardiac arrhythmias, anaphylactic reaction
- Increased incidence of icterus neonatorum
Pregnancy & Lactation
- Pregnancy category C
- Unknown excretion of Oxytocin in human milk
Precautions & Warnings
- Inducin should be used with care due to varying uterine sensitivity
- Smallest effective dose should be determined for each patient
- Should only be used with adequate medical supervision
- Fetal heart rate and maternal blood pressure should be determined
- Used with care if scars of the uterus and cervix uteri exist
Overdose Effects
- Excess Inducin may cause violent uterine contractions
- Leading to fetal bradycardia, arrhythmias, extensive laceration of soft tissues, uterine rupture, and possibly fetal or maternal death

Reconstitution
- Potency maintained for at least 24 hours when diluted with specific infusion fluids
- Direct injection into infusion line possible with certain infusion fluids
